Spacing recognition in the RXR/TR heterodimer


The RXR/TR (thyroid receptor) receptor heterodimer recognizes only the DR4 site, as seen in 2NLL. RXR binds "upstream," as it usually does. Protein-protein contacts are formed between loop of the TR and the second ZN-binding module of RXR. The T-box of RXR is disordered and it is not in position to interact. The extended C-terminal "A helix" of TR interacts with DNA, conferring additional binding affinity and specificity.
